Can someone tell me if a house needs to have a minimum size lot in order to build a second story addition?

What you are asking would be covered by the local zoning and planing commision, if you have one. Often they also have restrictions on maximum height.

Also your development might have C&R (covenants and restrictions) and/or a home owners association that will also control this.
