Hello

I have 1x6 T&G down acting as first subfloor but I want to put down 3/4 t&g plywood ontop is there a specific way to lay plywood or a certain side up?

I have beams 4 feet on center no joists...the 1x6 is down in position already

Help?

You will want to lay the long axis of the plywood perpendicular to the beams.

You are aware that the floor framing is not even close to standard building practice, & that with two ¾ inch subfloors it will be spongy underfoot.

To stiffen the floor, a third ¾ layer is advised, with gluing & screwing of all layers.
