My garage is approximately 21x21 and it has a 11x12 ft. office that was put in by previous owner. I would like to make another large room besides the office. How do I begin?

You could just lay out the area on paper and figure where it would be best located.
Then start framing your floor plan starting with a bottom plate, if on concrete you need to use a pressure treated bottom plate, frame the walls with the door included (2'8"x6'8" door is most common and requires an R.O. of 2'10"x6'9-10" depending on flooring)and cap with a double top plate.
