I built a screen for my projector on the media room and decided to put 2 last coats of semigloss,needless to say it looks awful, can I paint it over with flat paint just like that or do I need to sand it?

For the best results, I'd recommend buying special movie screen fabric and create a screen frame out of pine. Use the frame to stretch the fabric evenly (not too tight!) Paint the pine frame flat black. Total cost of around $100 and I think you'll be delighted with the quality of the picture -- much, much better than paint.
