Cold spots

Cold spots are a result of too wide a spacing of the tubing, or 'dead' tubes where water does not flow enough.

A reverse-flow pattern where the tubes go out through the floor and the return tubes come back parallel to the incoming tubes can drop off enough heat on the way in to not leave enough to heat above those cooler tubes coming back in the same floor, so the effect is like having wider spaced tubes.

Some more description of how the tubing was installed would give more answers. Gypcrete is good; but would not make a noticeable difference due to heat transfer. Concrete or gypcrete depth of floor holding the tubing has an effect.
