insulating a concrete floor

I am installing a wood subfloor on my concrete floor slab (poured 20 years ago). I was told to put polythene on top of the concrete, then the 2X4's on edge on top of that, then plywood flooring. What, if anything, can I put down for insulation, to keep coldness off the floor?
Fibreglass batting or styrofoam sheets, or nothing at all.
