I would not try to limit the migration of moisture either from the walls or into the walls. You want the walls to breathe in order to stay dry.
Space your 2x4 walls away from the basement walls 1/2 to 1 inch so they are not in direct contact with the block. Use treated lumber for the sill plate. Use rigid foam such as styrofoam, do not use fiberglass.
If you feel you must use plastic, put it against the block wall ONLY. If you have 2 layers, moisture will become trapped between.
By the way, the floor to wall is probably sealed to help prevent radon infusion.
