Did you consider cutting back the sewer line enough to install a new closet ring flush with the finished floor? By the way, how did you lose 4" floor height on a concrete floor? Has your work dropped the entire floor 4" below the level of the adjacent room? If that is the situation, it seems that you might want to build up the entire floor with 2 X 4 sleepers for a level flooring system. But back to your original question of attaching 2 X 4s to the concrete floor, since you are standing then on edge, it will be tough to screw or shoot anchors long ways through the framing members. However, you might build your frame, then place 2 x 4s inside the frame flat on the floor using Tapcon screws or power driven anchors. Then screw the raised floor frame into the edge of the 2 x 4s attached to the concrete floor.
