in my case there was extensive mildew on all the backer board, most of which was greenboard. I removed all the tile and backer behind the tile and installed wonderrboard backer and retiled with new tile (old tile was the vintage salmon (UGH) color) if the only damage is in the vicinity of the old repair, you may get by with just removing the affected tile, clean out the mess and retile. if there is mildew or mold, remove the backer if greenboard and replace with a backerboard. you may be able to kill the spores, but I think you are better off replacing any damaged or moldy backer.
