Do you have a crawl space under the house? If so, this is where you need to begin your exploring. Water runs down eventually, so the black stain of the leak will be visible under that room.
Many bathroom leaks originate with the shower or the comode that is leaking and has undermined the floor under the comode. An awl or ice pick doing some careful probing will also find the mushy area.
